چکیده

RNA(i) interference is a gene silencing mechanism triggered by double-stranded (ds)RNA, which promises to contribute species-specific insect pest control strategies. The first step toward the application of RNAi as an insecticide enable efficient upon dsRNA oral delivery. desert locust, Schistocerca gregaria devastating agricultural pest. While this species responsive delivered intra-hemocoelic injection, it refractory orally dsRNA. In study, we evaluated capacity five cell-penetrating peptides (CPPs) bind long and protect from locust midgut environment. We then selected CPP EB1 for further in vivo studies. EB1:dsRNA complexes failed induce feeding. Interestingly, observed that injection small-interfering (si)RNAs does not result response, but response can be obtained injecting EB1:siRNA complexes. also protected siRNAs degradation activity. However, well triggering when fed. Our findings highlight complexity dsRNA/siRNA-triggered emphasize multifactorial nature insects. study stresses importance studies comes dsRNA/siRNA delivery systems.

The vibrational startle response of the desert locust Schistocerca gregaria.

Substratum vibrations elicit a fast startle response in unrestrained quiescent desert locusts (Schistocerca gregaria). The response is graded with stimulus intensity and consists of a small, rapid but conspicuous movement of the legs and body, but it does not result in any positional change of the animal. With stimuli just above threshold, it begins with a fast twitch of the hindlegs generated ...

Ecdysone 20-mono-oxygenase in the desert locust, Schistocerca gregaria.

The enzyme catalysing the hydroxylation of ecdysone to 20-hydroxyecdysone, ecdysone 20-mono-oxygenase (EC 1.14.99.22), was investigated in the Malpighian tubules of fifth-instar locusts, Schistocerca gregaria. Enzyme activity was optimal at 35 degrees C and pH 6.8-8.0. Under these conditions the mono-oxygenase exhibited an apparent Km for ecdysone of 7.1 X 10(-7) M, a maximal specific activity ...

Dynamic flight stability in the desert locust Schistocerca gregaria.

Here we provide the first formal quantitative analysis of dynamic stability in a flying animal. By measuring the longitudinal static stability derivatives and mass distribution of desert locusts Schistocerca gregaria, we find that their static stability and static control responses are insufficient to provide asymptotic longitudinal dynamic stability unless they are sensitive to pitch attitude ...

Sulfakinins reduce food intake in the desert locust, Schistocerca gregaria.

In vertebrates, the peptides cholecystokinin (CCK), neuropeptide Y, galanin, and bombesin are known to be involved in the control of food intake. We report here that insect sulfakinins, peptides which display substantial sequence similarities with the vertebrate gastrin/CCK peptide family, significantly inhibit food uptake in fifth instar nymphs of the locust, Schistocerca gregaria. Upon inject...

Associative olfactory learning in the desert locust, Schistocerca gregaria.

Locusts can learn associations between olfactory stimuli and food rewards, and use the acquired memories to choose between foods according to their nutrient requirements. They are a model system for both the study of olfactory coding and insect nutritional regulation. Previous studies have used operant paradigms for conditioning freely moving locusts, restricting the study of the neural mechani...
